Amelioration of metformin-induced hypothyroidism by Withania somnifera and Bauhinia purpurea extracts in Type 2 diabetic mice

An investigation was carried out to reveal the possible ameliorative role of two plant extracts on an antidiabetic drug-induced hypothyroidism in Type 2 diabetic animals. Dexamethasone (1.0 mg/kg, i.m.) administration caused hyperglycemia with a parallel increase in renal lipid peroxidation (LPO), relative risk ratio (RR), and the concentrations of serum insulin; total cholesterol (TC); low-density lipoprotein cholesterol (LDL-C); very low-density lipoprotein cholesterol (VLDL-C) and triglycerides (TG). It decreased serum triiodothyronine (T3), thyroxine (T4) and high-density lipoprotein cholesterol (HDL-C) levels as well as renal superoxide dismutase (SOD); catalase (CAT) and reduced glutathione (GSH) content. Administration with metformin (150 mg/kg, orally) to diabetic animals further reduced circulating T4 level and caused severe hypothyroidism. It also reduced renal LPO, RR, serum concentrations of insulin; glucose and LDL-C with a parallel increase in cellular antioxidants. While oral administration with either Withania somnifera (1.4 g/kg) or Bauhinia purpurea (2.5 mg/kg) extract along with dexamethasone and metformin elevated the concentrations of circulating T3 and T4 to euthyroid level. The plant extracts also corrected RR ratio and serum concentration of lipids. The findings of the present study, for the first time, reveal that the evaluated plant extracts have a potential to ameliorate metformin-induced hypothyroidism in Type 2 diabetic subjects.

In conclusion, BVBE exhibited significant antihyperglycemic effects which may be attributed to increased glucose metabolism.


Antidiabetic activity of Bauhinia forficata extracts in alloxan-diabetic rats

The antidiabetic activity of aqueous, ethanolic and hexanic extracts of Bauhinia forficata was investigated in a model of alloxan-induced diabetes in rats. The biochemical parameters studied were: plasma glucose, serum triglycerides, cholesterol, high density lipoprotein (HDL), and low density lipoprotein (LDL). Extracts were administered daily for 7 d at doses of 200 and 400 mg/kg, p.o., 48 h after alloxan injection (60 mg/kg, i.v.). The alloxan-diabetic rats showed significant reductions in plasma glucose, triglycerides, total cholesterol and HDL-cholesterol after treatment with the extracts and glibenclamide (used as standard) as compared to the diabetic controls. Levels of LDL were not altered. In conclusion, our results showed that the plant extracts when administered by gavage may reduce glucose, triglycerides, total cholesterol and HDL-cholesterol levels. These results suggest the validity of the clinical use of B. forficata in the treatment of diabetes mellitus type II.

Hypoglycaemic and coronary risk index lowering effects of Bauhinia thoningii in alloxan induced diabetic rats

BACKGROUND: Disease, one of humanity's greatest adversaries, has in recent times showed an intimidating increase in numerical and pathological strength. This stretched the available medications to the limit thereby necessitating the need for the discovery of new and alternative medications to combat the menace of disease. Diabetic mellitus is one disease condition for which ideal synthetic drugs are yet to be discovered. To this end, pharmaceuticals are looking in the direction of medicinal plants. OBJECTIVE: This work aimed at screening Bauhinia thoningii (leaves) for its hypoglycaemic effect. The effect of the extract on lipid profile as a Coronary Risk Index (CRI) was also evaluated. METHODS: Aqueous crude extract of the plant was administered orally to alloxan induced diabetic rats and fasting blood glucose monitored over a period of 7 days. Blood samples collected from the rats were assayed for full lipid profile and the CRI calculated. RESULTS: Bauhinia thoningii caused 81.37% reduction in blood glucose of the experimental animals over a period of 7 days from an initial 365 mg/dl to 68 mg/dl. The plant extract was also observed to have the capacity to ameliorate diabetic complications like cardiovascular disorders. The extract reduced the Low Density lipoprotein (LDL) and reduced the CRI. CONCLUSIONS: Results from this study confirmed the hypoglycaemic efficacy of the extract and ability to ameliorate coronary diabetic complications. Further study is required to purify the plant extract to identify the fraction(s) that are responsible for the hypoglycaemic effects observed. This will also help to isolate the active components and elucidate the likely mechanism of action of the plant extract.
